<abstract xmlns="http://www.w3.org/1999/xhtml">

<p>This year we are celebrating the 200th anniversary of the birth of Louis Pasteur, one of the fathers of microbiology. Interestingly, the time when Pasteur disproved the doctrine of „spontaneous generation” and announced the “germ theory of disease” coincides with the discovery of <italic>Cryptococcus neoformans</italic> and its role in cryptococcosis. Today, only in the realm of guesswork can remain the correct answer to the question „whether the observed parallelism of these events was accidental?” or „whether Pasteur’s discoveries constituted a solid foundation of the research on the etiological factors of cryptococcosis?”. Until recently, it might seem that all major virulence factors of pathogenic fungi of the <italic>Cryptococcus</italic> species complex have been thoroughly described. Meanwhile, the simultaneous publication in 2018 of three <italic>in vitro</italic> protocols for the induction of Titan cells, also known as giant cells, opened up new possibilities for research on the relatively uncharacterized virulence factor that is crucial for <italic>Cryptococcus</italic> spp. Research on the titanization process makes us realize how little we know about the virulence factors of these fungi, and how much more can be improved in the context of the treatment and prevention of cryptococcosis. The following review is not only a historical outline of research on <italic>Cryptococcus</italic> spp. and cryptococcosis, but also synthetically describes the virulence factors of these basidiomycetous yeasts, with particular emphasis on the titanization process. The phenomenon of titanization as a process of a specific morphological transformation, like Titan cells, are completely new terms in Polish literature, which will be introduced to readers here. We live in a post-antibiotic era where the lack of effective and non-toxic drugs affects patients all over the world. Specifically, the availability of only fluconazole, amphotericin B and flucytosine in therapy of cryptococcosis constitutes a significant limitation. For this reason, research on the virulence factors of <italic>Cryptococcus</italic> spp. will allow to find new effective antimycotics, including inhibitors of the titanization process.</p>

<p>W bieżącym roku obchodzimy 200-setną rocznicę urodzin Ludwika Pasteur’a, jednego z ojców mikrobiologii. Od chwili obalenia przez niego „teorii samorództwa” i ogłoszenia „zarazkowej teorii chorób” upłynęło niemalże tyle samo czasu, co od odkrycia <italic>Cryptococcus neoformans</italic> i opisania jego roli w patogenezie kryptokokozy. Dziś w sferze domysłów może pozostawać właściwa odpowiedź na pytanie: Czy obserwowana równoległość zdarzeń była przypadkowa, czy może raczej odkrycia Pasteur’a stanowiły solidny fundament dla badań nad czynnikami etiologicznymi kryptokokozy? Do niedawna jeszcze mogło się wydawać, że wiemy już bardzo dużo o czynnikach wirulencji patogennych grzybów z kompleksu gatunków <italic>Cryptococcus.</italic> Tymczasem, równoczesna publikacja w 2018 roku trzech protokołów indukcji komórek Tytan <italic>in vitro</italic> ukazała nowe możliwości badań nad kluczowym dla <italic>Cryptococcus</italic> spp. czynnikiem wirulencji. Badania nad procesem tytanizacji uświadamiają nam jak mało jeszcze wiemy na temat czynników zjadliwości tych grzybów, a zarazem ile jeszcze można zrobić w kontekście leczenia i profilaktyki kryptokokozy. Niniejsza praca stanowi nie tylko rys historyczny badań nad <italic>Cryptococcus</italic> spp. i kryptokokozą, ale także w sposób syntetyczny opisuje czynniki wirulencji tych drożdży podstawkowych ze szczególnym uwzględnieniem procesu tytanizacji. Zjawisko tytanizacji jako proces swoistej transformacji morfologicznej, podobnie jak komórki Tytan są terminami zupełnie nowymi w literaturze polskiej, które na łamach niniejszej pracy zostaną przybliżone czytelnikom. Nie ulega wątpliwości, że żyjemy w erze post-antybiotykowej gdzie brak skutecznych i nietoksycznych leków dotyka pacjentów na całym świecie. Podobnie, dostępność w terapii kryptokokozy jedynie flukonazolu, amfoterycyny B i flucytozyny nie napawa optymizmem i stanowi znaczne ograniczenie. Z tego względu badania nad czynnikami wirulencji <italic>Cryptococcus</italic> spp. pozwolą znaleźć nowe skuteczne antymykotyki, w tym inhibitory procesu tytanizacji.</p>
