Several commercial sources of tobacco-derived nicotine (TDN) and synthetic nicotine (SyN) and a variety of e-cigarette liquids employing either TDN or SyN have been evaluated to determine the enantiomer distributions of R- and S-nicotine and R- and S-nornicotine by chiral supercritical fluid chromatography (chiral-SFC) with UV diode array detection (DAD-UV). The data generated are used to test the mismatched <italic>vs</italic>. matched hypothesis of C<sc>heetham</sc> <italic>et al.</italic> as a means to distinguish products containing TDN from products with SyN.
Two sets of experiments were conducted in this study. The first experiment was conducted on a series of 11 commercial nicotine samples (three characterized as tobacco-derived and eight characterized as synthetic nicotine). The commercial nicotine samples were either from a tobacco-derived nicotine (TDN) source or were synthetic nicotine (SyN). Some of the commercial nicotine samples were nicotine salts. The second experiment was conducted on e-liquids from a set of 11 e-cigarettes. The nicotine in the e-liquids was either from TDN or SyN. The e-liquid samples were differentiated based on the advertised information on the internet or from printed information on the e-cigarette packaging.
None of the three commercial TDN samples in the first experiment could be unequivocally characterized as coming from a tobacco source. Five of the eight commercial SyN samples were correctly characterized as SyN based on the matched <italic>vs</italic>. mismatched nicotine and nornicotine hypothesis of C<sc>heetham</sc> <italic>et al.</italic>
In the second experiment, none of the e-liquids characterized as being from TDN sources could be unequivocally characterized as coming from a tobacco source. All of the e-liquids characterized as being from SyN sources were either characterized as equivocal or of uncertain origin based on the matched <italic>vs</italic>. mismatched nicotine and nornicotine hypothesis of C<sc>heetham</sc> <italic>et al.</italic>
These sets of experiments represent an excellent example of the difficulty that the United States Food and Drug Administration is having in trying to determine if TDN or SyN is being used in tobacco products. Even highly advanced chromatographic methods such as chiral-SFC were not able to unequivocally distinguish products with TDN from products with SyN 100% of the time.
Other analytical methods such as 14C quantitation of nicotine samples by accelerator mass spectrometry offer a more reliable determinate of nicotine source (TDN <italic>vs</italic>. SyN) and can be used to identify misbranded products labelled as containing SyN, even though this methodology is more expensive and offered in limited locations.

Qualitative and Quantitative Analyses of the Enantiomers of Nicotine and Related Alkaloids Employing Chiral Supercritical Fluid Chromatography in Commercial Nicotine Samples and in E-Cigarette Products

Es erfolgte eine Untersuchung mehrerer im Handel erhältlicher Quellen von aus Tabak gewonnenem Nikotin (TDN, <italic>tobacco-derived nicotine</italic>) und synthetischem Nikotin (SyN) sowie einer Auswahl von Liquids für E-Zigaretten, die jeweils TDN- oder SyN enthielten. Mithilfe der chiralen überkritischen Flüssigkeitschromatographie (chirale SFC, <italic>supercritical fluid chromatography</italic>) mit UV-Diodenarray-Detektor (DAD-UV) die Verteilung der Enantiomere R-und S-Nikotin sowie R- und S-Nornikotin bestimmt werden sollte. Die so erhobenen Daten wurden verwendet um die Match-Mismatch-Hypothese zu prüfen, die von C<sc>heetham</sc> <italic>et al</italic>. zur Unterscheidung von Produkten mit TDN von solchen mit SyN vorgeschlagen wurde.
Im Rahmen dieser Studie wurden zwei Versuchsreihen durchgeführt. Der erste Versuch erfolgte an 11 handelsüblichen Nikotinproben (wovon drei als aus Tabak gewonnen und acht als synthetisches Nikotin gekennzeichnet waren).
Bei einigen der handelsüblichen Proben handelte es sich um Nikotinsalze. Der zweite Versuch wurde mit E-Liquids aus 11 E-Zigaretten durchgeführt. Bei dem Nikotin in den E-Liquids handelte es sich entweder um TDN oder SyN. Die Unterscheidung der E-Liquid-Proben erfolgte auf der Grundlage der verfügbaren Angaben im Internet bzw. anhand der auf der Verpackung der E-Zigaretten aufgedruckten Informationen. Keine der handelsüblichen TDN-Proben konnte im ersten Experiment eindeutig als aus einer Tabakquelle gewonnen bestimmt werden. Fünf der acht handelsüblichen SyN-Proben waren, ausgehend von der Match-Mismatch-Hypothese von C<sc>heetham</sc> <italic>et al</italic>. für Nikotin und Nornikotin, korrekt als SyN gekennzeichnet. Im zweiten Experiment konnte keines der E-Liquids, das als aus einer TDN-Quelle gewonnen gekennzeichnet war, eindeutig als aus einer Tabakquelle gewonnen bestimmt werden. Sämtliche E-Liquids, die als aus SyN-Quellen gewonnen gekennzeichnet waren, wurden, ausgehend von der ‘Match-Mismatch-Hypothese’ von C<sc>heetham</sc> <italic>et al</italic>. für Nikotin und Nornikotin, entweder als nicht eindeutig oder als aus unklarer Herkunft eingestuft.
Diese Versuchsreihen sind ein exzellentes Beispiel für die Schwierigkeiten, denen die US-Arzneimittelbehörde FDA bei dem Versuch begegnet, festzustellen, ob in Tabakerzeugnissen TDN oder SyN-Nikotin verwendet wird. Selbst mithilfe hochmoderner chromatographischer Methoden wie der chiralen SFC war es nicht immer möglich, Produkte mit TDN eindeutig von Produkten mit SyN-Nikotin zu unterscheiden.
Andere analytische Methoden wie die C14-Methode zur Quantifizierung von Nikotinproben mithilfe einer Beschleuniger-Massenspektrometrie würden eine zuverlässigere Bestimmung der Nikotinquelle (TDN oder SyN) ermöglichen und könnten eingesetzt werden, um Produkte zu ermitteln, die fälschlich als mit SyN deklariert wurden. Diese Methoden sind allerdings teurer und stehen nicht überall zur Verfügung.

Plusieurs sources disponibles dans le commerce de nicotine extraite du tabac et de nicotine de synthèse ainsi qu’un éventail de liquides pour cigarettes électroniques à base de nicotine extraite du tabac ou de nicotine de synthèse furent évalués dans le but de déterminer les répartitions des isomères optiques R- et S-nicotine et R- et S-nornicotine par le biais d’une chromatographie en phase supercritique chirale (CPS chirale) couplée à l’usage d’un détecteur UV à barrette de diodes. Les données récoltées furent utilisées pour mettre à l’épreuve l’hypothèse ‘mismatched <italic>vs</italic>. matched’ de C<sc>heetham</sc> et consorts servant à distinguer les produits contenant de la nicotine extraite du tabac des produits contenant de la nicotine de synthèse.
Deux séries d’expériences furent menées lors de la présente étude. Les premiers essais furent réalisés sur une série de 11 échantillons de nicotine disponible dans le commerce (trois caractérisés comme étant extraits du tabac et huit caractérisés comme de la nicotine de synthèse). Les échantillons de nicotine disponible dans le commerce contenaient soit de la nicotine extraite du tabac, soit de la nicotine de synthèse. Certains des échantillons de nicotine disponible dans le commerce étaient des sels de nicotine. Les deuxièmes essais furent menés sur des liquides sélectionnés parmi un éventail de 11 cigarettes électroniques. La nicotine présente dans les liquides était soit extraite du tabac, soit un composé de synthèse. Les échantillons de liquides furent différenciés sur la base des informations publiées sur l’Internet ou imprimées sur les emballages des cigarettes électroniques.
Pendant la première des séries d’expériences aucun des trois échantillons de nicotine extraite du tabac et disponible dans le commerce ne put irréfutablement être caractérisé comme provenant d’une source de tabac. Cinq des huit échantillons de nicotine de synthèse disponible dans le commerce furent correctement caractérisés comme de la nicotine de synthèse sur la base de l’hypothèse ‘matched <italic>vs.</italic> mismatched’ de C<sc>heetham</sc> et consorts pour la nicotine et la nornicotine.
Pendant la seconde des séries d’expériences aucun des liquides pour cigarettes électroniques identifiés comme provenant d’une source de nicotine extraite du tabac ne put irréfutablement être caractérisé comme provenant d’une source de tabac. Tous les liquides identifiés comme provenant d’une source de nicotine de synthèse furent caractérisés soit comme équivoques, soit d’origine incertaine sur la base de l’hypothèse ‘matched <italic>vs.</italic> mismatched’ de C<sc>heetham</sc> et consorts pour la nicotine et la nornicotine.
Ces séries d’expériences apportent un excellent exemple de la difficulté rencontrée par la ‘United States Food and Drug Administration’ dans ses tentatives de déterminer la présence de nicotine extraite du tabac ou de nicotine de synthèse dans les produits de tabac. Même des méthodes recourant à une chromatographie de pointe telles que la CPS chirale ne permirent pas de distinguer, sans équivoque et à chaque essai, les produits contenant de la nicotine extraite du tabac des produits contenant de la nicotine de synthèse.
D’autres méthodes analytiques telles que l’analyse chimique quantitative au carbone 14 des échantillons de nicotine à l’aide d’une spectrométrie de masse par accélérateur ouvrent des pistes plus fiables de détermination de la source de nicotine (tabac ou synthèse) et peuvent être appliquées pour identifier les produits mal étiquetés contenant prétendument de la nicotine de synthèse, même si cette méthodologie s’avère plus coûteuse et n’est pas disponible partout.
