<abstract xmlns="http://www.w3.org/1999/xhtml">

The present study evaluated the <italic>in vitro</italic> extraction of benzo[<italic>a</italic>]pyrene (BaP) from moist snuff into water and into artificial saliva. A similar, previous study evaluated the levels of BaP that remained in the moist snuff after the extraction but did not measure the levels of BaP in the water or saliva extract. The previous study showed that the remaining levels of BaP in the solid material were between 96.3% and 109.6% relative to the initial level of BaP, when the snuff was washed with water and between 99.4% and 108.3% from the initial level of BaP, when the snuff was washed with both saliva and water. Nine moist snuff samples (eight being the same brands as evaluated in the previous study) were analyzed in the present study. Several improvements were made compared to the previous study regarding the extraction conditions. The extraction was performed for 1 h at 37 °C, using a mechanical agitator.
The previous study used a commercially available artificial saliva which had an adjusted pH but did not contain enzymes or salts. This saliva was replaced with complete artificial saliva containing salts, mucin and enzymes. The results indicated that the level of BaP extracted in 100 mL water from 5 g of moist snuff at 37 °C ranged between 1.0% and 1.7% of the initial level present in tobacco. For artificial saliva, the extracted level of BaP was between 2% and 3.9% from the initial level, depending on the moist snuff brand. Although the BaP level extracted from the moist snuff with artificial saliva remained very low, the surfactant character of artificial saliva increased BaP extraction relative to water by a factor of approximately two. This study supports the previous reported finding that the vast majority of BaP in moist snuff is not extracted in water or artificial saliva.

In der vorliegenden Studie wurde die <italic>in vitro</italic>-Extraktion von Benzo[<italic>a</italic>]pyren (BaP) aus Moist Snuff (feuchtem Schnupftabak) mit Wasser und künstlichem Speichel untersucht. In einer ähnlichen, früheren Studie war zwar der Restgehalt an BaP im feuchten Schnupftabak nach Extraktion untersucht worden, es wurde jedoch nicht der BaP-Gehalt im Wasser bzw. im Speichelextrakt bestimmt. In der früheren Studie konnte gezeigt werden, dass die Menge an BaP in extrahiertem Moist Snuff zwischen 96,3% und 109,6% im Verhältnis zur Ausgangsmenge lag, wenn der Schnupftabak mit Wasser behandelt wurde, sowie zwischen 99,4% und 108,3%, wenn der Schnupftabak mit Wasser und Speichel behandelt wurde. In der vorliegenden Studie wurden nun neun Proben von Moist Snuff untersucht (bei acht Proben handelte es sich um dieselben Marken, die in der früheren Studie untersucht wurden). Im Vergleich zur vorherigen Studie wurden bei den Extraktionsbedingungen mehrere Verbesserungen vorgenommen. Die Extraktion erfolgte für eine Stunde bei 37 °C unter Einsatz eines mechanischen Rührers. In der früheren Studie war handelsüblicher künstlicher Speichel verwendet worden, der zwar einen angepassten pH-Wert hatte, jedoch keine Enzyme oder Salze enthielt. Dieser Speichel wurde nun durch einen vollständigen künstlichen Speichel ersetzt, der Salze, Muzin und Enzyme enthielt. Die Ergebnisse zeigten, dass die Menge an BaP, die durch Extraktion in 100 mL Wasser aus 5 g Moist Snuff bei 37 °C zu finden war, zwischen 1,0% und 1,7% der im Tabak vorliegenden Ausgangskonzentration lag. Bei künstlichem Speichel lag, in Abhängigkeit von der Marke des Tabaks, der extrahierte BaP-Gehalt zwischen 2,0% und 3,9% der im Tabak zu findenden Ausgangskonzentration. Die mit künstlichem Speichel aus dem Tabak extrahierten BaP-Konzentrationen waren sehr niedrig. Der Tensidcharakter des künstlichen Speichels erhöht jedoch die Extraktion von BaP im Verhältnis zu Wasser ca. um den Faktor zwei. Mit der vorliegenden Studie wird das Ergebnis der früheren Studie untermauert, wonach der überwiegende Anteil an BaP in Moist Snuff nicht mit Wasser oder künstlichem Speichel extrahiert wird.

La présente étude évalua l’extraction <italic>in vitro</italic> du benzo[<italic>a</italic>]pyrène (BaP) présent dans le tabac à priser humide à l’aide d’eau et de salive artificielle. Une étude précédente mais similaire avait évalué les niveaux de BaP qui subsistent dans le tabac à priser humide après extraction mais n’avait pas mesuré les niveaux de BaP dans les extraits de salive ou d’eau. L’étude précédente avait démontré que les teneurs résiduelles de BaP dans la matière solide oscillaient entre 96,3% et 109,6% par rapport à la teneur initiale en BaP après le lavage du tabac à priser à l’eau et entre 99,4% et 108,3% de la teneur initiale en BaP après le lavage du tabac à priser à la salive et à l’eau. Neuf échantillons de tabac à priser humide (huit provenant des mêmes marques que celles analysées dans l’étude précédente) furent analysés dans la présente étude. Comparativement à l’étude précédente, plusieurs améliorations furent apportées quant aux conditions d’extraction. L’extraction fut réalisée durant 1 heure à 37 °C, à l’aide d’un agitateur mécanique. L’étude précédente avait eu recours à une salive artificielle disponible sur le marché affichant un pH ajusté mais dépourvue d’enzymes ou de sels. Cette salive fut remplacée par une salive complètement artificielle contenant des sels, de la mucine et des enzymes. Les résultats indiquèrent que la teneur en BaP extrait dans 100mL d’eau à partir de 5g de tabac à priser humide portés à 37 °C oscillait entre 1,0% et 1,7% de la teneur initiale présente dans le tabac. Dans le cas de la salive artificielle, la teneur en BaP extraite oscillait entre 2,0% et 3,9% de la teneur initiale, selon la marque de tabac à priser. Bien que la teneur de BaP extraite du tabac à priser humide dans la salive artificielle demeure très faible, les propriétés surfactantes de la salive artificielle augmente, d’un facteur proche de deux par rapport à l’eau, l’extraction de BaP. Cette étude étaye la conclusion précédemment présentée selon laquelle la grande majorité du BaP présent dans le tabac à priser humide n’est pas extraite dans l’eau ou la salive artificielle.
