During the last 35 years technologies and hardware for the separation of enantiomers have advanced tremendously. Today, complete resolution of the enantiomers of nicotine and related compounds can effectively be attained within a few seconds and most often less than 10 minutes. In some unique cases, enantiomeric separation for nicotine-related compounds having a wide variety of functional groups has been demonstrated. Particularly, the successful wedding of short HPLC and SFC columns containing very small particle size materials bonded to chiral stationary phase materials coupled with information-rich detectors like electrospray ionization mass spectrometry/mass spectrometry has facilitated this remarkable transition. Taken collectively the HPLC and SFC technologies will allow for very rapid (seconds), reliable, and reproducible (% RSD routinely &lt; 5%) for nicotine and related alkaloid enantiomers.

Qualitative and Quantitative Analysis of Nicotine, Nicotine Derivatives, and Nicotine-Related Alkaloid Optical Isomers: A Review

In den letzten 35 Jahren hat es bei der Technik und den Instrumenten zur Trennung von Enantiomeren beträchtliche Fortschritte gegeben. Aktuell kann eine vollständige Trennung der Enantiomere von Nikotin und verwandten Verbindungen effektiv innerhalb von wenigen Sekunden bzw. meist in unter 10 Minuten erfolgen. In Einzelfällen konnte gezeigt werden, dass eine Enantiomerenseparation bei mit Nikotin verwandten Verbindungen mit einer großen Vielzahl funktioneller Gruppen möglich ist. Erleichtert wurde diese bemerkenswerte Entwicklung insbesondere durch die erfolgreiche Kombination von kurzen Säulen für die Hochdruckflüssigkeitschromatographie (HPLC) und die überkritische Flüssigkeitschromatographie (SFC) aus Materialien mit sehr kleinen Partikelgrößen, gebunden an chirale stationäre Phasenmaterialien und gekoppelt mit Detektoren wie Elektrospray-Ionisations-Massenspektrometrie/Massenspektrometrie. Die kombinierte Anwendung der HPLC- und SFC-Technologie kann daher eine Trennung der Enantiomere von Nikotin und verwandten Alkaloiden ermöglichen, die sehr schnell (innerhalb von Sekunden), zuverlässig und reproduzierbar (relative Standard-abweichung RSD routinemäßig &lt; 5%) ist.

De nos jours, une résolution complète des isomères optiques de la nicotine et de ses composés apparentés peut, dans les faits, être obtenue en l’espace de quelques secondes et habituellement en moins de 10 minutes. Dans certains cas isolés, il a été démontré qu’il était possible de séparer des énantiomères des composés apparentés à la nicotine ayant des groupes fonctionnels très variés. Cette remarquable avancée fut notamment facilitée par l’utilisation combinée réussie de colonnes courtes de la CLHP (chromatographie liquide haute performance) et de la CPS (chromatographie en phase supercritique), contenant des matériaux de particules de très petite taille liées à des matériaux en phase stationnaire chirale et couplées à de puissants capteurs d’information tels que la spectrométrie de masse et la spectrométrie de masse à ionisation par électro-nébulisation. Conjuguées, les technologie CLHP et CPS permettront un analyse séparative très rapide (de l’ordre de quelques secondes), fiable et reproductible (écart type de répétabilité habituellement inférieur à 5%) des énantiomères de la nicotine et des alcaloïdes apparentés.
