Abstrakt

Abstract

The aim of our work was to investigate the effect of amitriptyline, citalopram and venlafaxine on the heart during ischemic- reperfusion (l-R) injury. Amitriptyline prolonged both QRS complex and QTc interval duration; citalopram and venlafaxine prolonged only QTc interval duration. Amitriptyline worked most proarrhythmogenic, citalopram least; venlafaxine increased the heart rate during ischemia; however, prolonged QTc interval at the beginning of reperfusion was followed by serious dysrhythmias.

The incidence of dysrhythmias after administration of the antipsychotic olanzapine

Abstrakt

Abstract

We evaluated the effect of the antipsychotic olanzapine on electrical activity of rat hearts under conditions of ischemic- reperfusion injury. We focused on the prolongation of the corrected QT interval as a risk factor for the incidence of different types of dysrhythmias. Pretreatment with olanzapine showed prolongation of the corrected QT interval as well as increased incidence of dysrhythmias in following order: ventricular premature beats > bigeminies > trigeminies > salvos. We also observed an increase in the frequency of episodes of ventricular tachycardia of about 64% and the average duration of ventricular tachycardia was more than doubled under the conditions of the ischemic-reperfusion injury.

Voluntary exercise and testosterone therapy caused increase in percentage of Myh6 and expression of oxidative stress marker Cybb in left ventricles of rats

Abstrakt

Abstract

Aim: The aim of this study is to identify a possible damage to heart ventricles caused by supraphysiological doses of testosterone, voluntary physical activity or their combination.

Methods: In the 8-week long experiment, 10-12 weeks old male Wistar rats were administered testosterone depot in dose of 100 mg/kg (TES, n = 15) or vehiculum (CON, n = 12) once a week subcutaneously. Next groups injected with testosterone (SPOTES, n = 12) or vehiculum (SPO, n = 12) were running in exercise wheels ad libitum. Gene expressions in left and right ventricles of the heart were measured by quantitative reverse transcription polymerase chain reaction method.

Results:ln left ventricles of the testosterone groups, we observed a mild but significant increase in the percentage of Myh6 myosin heavy chain isoform and higher expression of NADPH oxidase subunit Cybb (*p < 0.05).

Conclusions:Testosterone affected the expression of genes related to contractile apparatus and oxidative stress in the left ventricle but not in right ventricle of the heart of rats. The observed level of physical activity did not have a compelling effect on the expression of measured genes.

Effect of Chronic Social Stress on Endothelial Function of the Mesenteric Artery of Normotensive and Spontaneously Hypertensive Rats

Abstract

The aim of the work was to study the effect of chronic social stress induced by increased population density, “crowding stress” on blood pressure and endothelial function of arteries of male normotensive Wistar and spontaneously hypertensive (SHR) rats. Based on the results, we assume that social stress-induced reduction in endothelium-dependent relaxation and increased blood pressure of SHR rats is most likely associated with decreased bioavailability of nitric oxide.

Acute phase of depression treatment - current research and its perspectives on Jessenius Faculty of Medicine

Abstract

In the previous research at Jessenius Faculty of Medicine in Martin has been studied depression and its treatment from the aspects of the quality of life and functioning of the patients, as well as comparing the efficacy and safety of venlafaxine versus venlafaxine and olanzapine treatment. Last years have been studied the parameters of the autonomic nervous system in the context of depression. Our new work will build on these findings, during the exploration of efficacy and safety of a novel antidepressant vortioxetine in monotherapy and in combination with olanzapine. Since combination of vortixetine and olanzapine is not yet understood, it is unknown whether the effect of the treatment of depressive symptoms and associated problems will be stronger and faster compared to vortioxetine monotherapy.

Is atomoxetine effective in some comorbid mental disorders in ADHD?

Abstrakt

Abstract

Attention-Deficit/Hyperactivity Disorder (ADHD) is connected with high level of psychiatric comorbidity in paediatric population. Depressive disorder is common comorbid disorder co-existing with ADHD. Atomoxetine is worldwide approved for treatment of ADHD in paediatric population; in addition atomoxetine is effective and safe in treatment of some comorbid disorders in ADHD. Pharmacotherapy of depression is limited and residual symptoms are common. Fluoxetine is currently considered to be the gold standard of treatment of depression, but effectiveness of acute phase of treatment is not sufficient. Atomoxetine as a selective noradrenaline reuptake inhibitor or olanzapine as a multi receptors antagonist drug in combination with fluoxetine could be perspective augmented treatment strategy of depression just for their antidepressant effect. The aim of our following study is to evaluate and compare effectiveness and safety of monotherapy and combined/augmented therapy in acute phase of depression treatment in adolescence, as well as introduce complex modern research methodology of effectiveness and safety of treatment.

Development and validation of novel self-report questionnaire about the assessment of adolescents’ relation to medicines use and risk

Abstrakt

Abstract

Adolescents have a lack of experience, knowledge of medicines, which often leads to incorrect use of medicines. Slovak adolescents' attitude towards the use and risk of medicines is not known, and therefore, we prepared a questionnaire and then we evaluated the face, content (CVR i <1) and construct validity and reliability (Cronbach’s α = 0.85). The questionnaire is an appropriate tool to assess the quality of knowledge and experience of adolescents with drugs and the data could, on the introduction of measures, contribute to the rational use of medicines by adolescents in Slovakia.
