Analysis of the Essential Oil Compounds of Origanum Syriacum L.

Abstrakt

Abstract

Syrian oregano (Origanum syriacum L., Lamiaceae) is a very popular culinary herb that has been used through the ages in traditional medicine mainly in Lebanon and the Arab world. The subject of this study was the microscopic analysis of the herbal drug Origani syriaci folium, as well as the chemical analysis of its essential oil components. Plant materials originated from Arabsalim - South Lebanon and were collected in 2010. The presence of very dense clusters of multicellular covering trichomes (510 μm × 18.47 μm) and peltate secretory glands (∅ 87.95 μm) with eight cells were observed. The essential oil of Origani syriaci folium was isolated and quantified using hydrodistillation according to SPhC 1997. The content of essential oil was 95 ml/1000 g, i.e. 9.5% (V/m). The volatile constituents of Origani syriaci folium were qualitatively and quantitatively evaluated using SPME GC/MS. The two dominant constituents identified in Syrian oregano leaves were carvacrol (78.4%) and thymol (17.9%).

Synthesis and HPLC Enantioseparation of Derivatives of the 3-hydroxyphenylethanone

Abstrakt

Abstract

Within the framework of the study of the synthesis and high-performance liquid chromatography (HPLC) enantioseparation the series of 9 derivatives of 3-hydroxyphenylethanone was prepared by a well-tried method. The structure of the prepared compounds was confirmed on the basis of interpretation of the IR, UV, 1H NMR and 13C NMR spectra. An enantioseparation of prepared compounds was performed using HPLC on a native teicoplanin (Chirobiotic T) and the amylose tris (3,5-dimethylphenylcarbamate) (Chiralpak AD) chiral stationary phases, which is more suitable for the enantioseparation of all prepared compounds especially with heterocycles in the basic part of a molecule.

Acute Pain Treatment at Department of Traumatology

Abstrakt

Abstract

The aim of this retrospective study was to characterize the acute pain management in patients admitted to Department of Traumatology and to identify the efficacy of analgesic pharmacotherapy. The study involved 83 patients divided into two groups: 52 patients (63%) in Group 1 underwent operation; the remaining 31 patients (37%) in Group 2 had conservative treatment. The characteristics of the patients (diagnosis, analgesic therapy, age, comorbidities, drugs used during hospitalisation, analgesic efficacy) were obtained from electronic database and medical reports by the hospital. Therapy during the first seven days of hospitalisation and the recommended therapy after discharging home were analysed. Records of drug adverse reactions during the whole hospitalisation were monitored. Metamizol was the most frequently prescribed as a primal analgesic. Adequate analgesia solely by metamizol was reached in 20% of all patients; the remaining 80% required increased doses or combination with another analgesic. Metamizol was combined mainly with tramadol, morphine or pethidine. Our study proved metamizol to be an effective analgesic. No clinical manifestation of agranulocytosis was observed in this study.

Solubilisation of (+)-Usnic Acid in Aqueous Micellar Solutions of Gemini and Heterogemini Surfactants and their Equimolar Mixture

Abstrakt

Abstract

The solubilisation of natural compound, (+)-usnic acid, in micellar solutions of gemini (N,N’-didecyl-N,N,N’,N’-tetramethylethane-1,2-diyldiammonium dibromide) and heterogemini (decyl 2-[decyl(dimethyl)ammonio]ethylphosphate) surfactants and their equimolar mixture has been studied. The highest solubility was found for gemini surfactant. The relationship between synergism in surface properties of mixture of surfactants and their solubilisation properties is also discussed.

Molecular Docking Study on the Binding Mode of Cardioselective Phenoxyaminopropanol Blocker into β-adrenergic Receptor Subtypes

Abstrakt

Abstract

Structural understanding of subtype specific ligand-binding pocket variations and interactions of ligand with receptor may facilitate design of novel selective drugs. To gain insights into the subtype selectivity of β-blockers we performed flexible molecular docking study to analyze the interaction mode of cardioselective phenoxyaminopropanol blocker into the β1 and β2-adrenergic receptor. The binding site analysis reveals a strong identity between important amino acid residues and interactions with ligand in orthosteric catecholamine- binding pocket. The differences in the binding mode of selective ligand have been identified in the extracellular region of receptor subtypes.

Evaluation of Stability of Potential Betaadrenolytics in Various Solutions by thin Layer Chromatography and by Kinetics of Hydrolysis

Abstract

The aim of this work is the study of stability and kinetics of hydrolysis of the chosen compounds, derivatives of 2-hydroxy-3-[2-(4-methoxyphenyl)ethylamino]propyl-4- [(alkoxycarbonyl)amino]benzoates and 2-hydroxy-3-[2-(2-methoxyphenyl)ethylamino] propyl-4-[(alkoxycarbonyl)amino]benzoates with potential ultra-short beta-adrenolytic activity. The studied compounds are different in the position of the substituent on the benzene ring in the side chain as well as in the aromatic ring in position 4 with alkyl- (methyl- to butyl-) carbamate. Thin layer chromatography and UV-area spectrophotometry are used in order to establish the stability of these potential pharmaceuticals. The stability studies of the compounds were examined in acidic and alkaline media, in buffers and due oxidation at room and at elevated temperature chromatographically, and Rf values of incipient products and degradation products were detected. Kinetics of acid and base hydrolysis in various solutions at temperatures 80 °C and 100 °C were examined through UV-area spectrophotometry. Kinetic parameters such as rate constant k, half-life period t1/2 and usable life t90 were determined.

The Effect of Methamphetamine on Biotransformation of Ethanol: Pilot Study

Abstrakt

Abstract

Methamphetamine is one of the most popular recreational drugs in Central Europe and is often combined with ethanol. Various interactions between these two substances have been described including the influence of administered ethanol on biotransformation of methamphetamine. The aim of the present study was to describe the opposite effect - the influence of methamphetamine on biotransformation of ethanol in rats. Methamphetamine was administered for 10 days (10 mg/kg/day) i.p. and ethanol was delivered as an intragastric bolus (2 g/kg) on the10th day of experiment to both methamphetamine administered rats and control animals. The pharmacokinetic experiment on the whole animal was performed and plasma samples were drawn at the 40th, 120th, 210th and 300th minute after ethanol administration. Ethanol plasmatic levels reached significantly lower values in the 40th and 120th interval when compared to controls. Differences were insignificant in the last two intervals. Our results suggest that chronic methamphetamine administration induces ethanol biotransformation. We suppose that this effect is caused by induction of alcohol dehydrogenase metabolic activity or by allosteric interaction of methamphetamine and this enzyme. More studies have to be conducted to confirm or disprove our hypothesis.
